Polyclonal antibody with avidity to ROS used for determination for ROS in plasma

The results of a study to find the determination of ROS in plasma have been revealed.

Published in the Chemistry Central Journal 2011, the experiment used a polyclonal antibody with high avidity and to ROS.

It was then used in the development of highly sensitive ELISA for determination of ROS in plasma.

A number of eight-week old New Zealand white rabbits were immunised with the ROS-KLH conjugate and were then monitored by direct ELISA using ROS-BSA immobilised onto microwell plates as a solid phase.

Sera was collected from the rabbit with the highest antibody titer and avidity to ROS and the IgG fraction was isolated and purified through avidity chromatography on protein A column.

The conclusion revealed that the purified antibody showed high avidity to ROS; IC50= 0.4ng/ml, with the concentration of ROS in the sample justified by its capacity to stop the binding of the anti-ROS antibody to the immobilised ROS-BSA.
