    Improve your GLP-1 purification with YMC

    Glucagon-like peptide-1 (GLP-1) is a peptide hormone that can consist of up to 30 amino acids. It has several physiological properties that make it (and its functional analogues) a subject of intense investigation as a potential treatment for diabetes mellitus. Nevertheless, the purification of GLP-1 is a challenging task. Reversed-phase (RP) chromatography has established itself as the preferred purification method. With the right stationary phase, structural variants and even peptides with minimal sequence differences can be precisely separated.     Extraordinary stability: chemical and mechanical

    In general, changing the pH value to extreme conditions can be helpful for difficult purification tasks. Especially for the purification of GLP-1, alkaline buffers are very beneficial for the effective binding and separation. But high pH cannot be used with silica-based phases. YMC-Triart Prep is based on a hybrid silica material. Due to the cross-linking with ethylene bridges, it provides exceptional chemical stability and can also be used at high pH values. This is advantageous for many peptide purifications, and drastically increases the possible method set-ups.  Furthermore YMC-Triart Prep stationary phases enable reproducible results when using 100% aqueous conditions. This is particularly useful for polar peptides.

    Common cleaning-in-place (CIP) procedures with 0.1 M NaOH can be used to regenerate the column and restore chromatographic performance. The YMC-Triart Prep stationary phases are stable for more than 300 CIP cycles. With its robustness and long lifetime, this packing media reduces process costs enormously. In combination with the excellent chromatographic performance and high loadability, YMC-Triart Prep can contribute to higher productivity of the overall process.

    Maximise your resolution

    The material provides high resolution to achieve the required purity. Since the selectivity has the greatest influence on the resolution it is recommended to screen different stationary phases. It often makes sense to consider other modifications in addition to the standard C18 phase. YMC-Triart Prep offers a wide range of different selectivities for the purification of GLP-1. By selecting the right stationary phase, the productivity and efficiency of the entire process can be determined. YMC-Triart Prep is completely scalable from analytical to preparative scale.
