Analysis of Phospholipids by HPTLC

Phospholipid analysis challenges any chromatographic technique, mainly due to the nature of the analytes. Historically, TLC has been used and the reproducibility of results has been unreliable and unquantifiable. At CAMAG several parameters of the chromatographic process were investigated including chamber saturation, derivatization, plate activity, and consistency of HPTLC plates.
The good news for researchers is that High Performance Thin Layer Chromatography separation and quantification of sphingomylin, PC, PI, PS, PG, PE, cardiolipin, PA, lyso PC, PI, PS, PG and PE can be achieved. Crucially, for reproducible results the employed methodology must be strictly standardized with the developing chamber homogenously saturated. An automatic developing chamber, such as CAMAG’s ADC 2, with humidity control is ideally suited. The findings of this research presents measures to help keep experimental conditions under tight control, providing reliable quantitative results and bringing economic benefits to industry.
